Laser - ExcitedImmunofluorometricAssayof Prolactin , with Use of Antibodies
نویسنده
چکیده
We describe an immunofluorometric assay for prolactin based on lanthanide labelingof a monoclonalantibody and measuring time-resolved fluorescence. In this “sandwich”type assay, the label (Eu3) was bound to the second antibody by means of a simple, rapid method involving the anhydride of diethylenetriaminepentaacetic acid. To measure the photoluminescence of europium (or other lanthanides), we have developed a time-resolved fluorometer with a nitrogen laser as the pulsed excitation source. During the assay, the solid-phase antibody immobilized inside a polystyrene tube is incubated with the plasma sample and the second antibody in a one-step procedure. Results for 67 human plasmas correlated well (r = 0.98) with those by an immunoradiometricmethod.

Use of immunoaffinity chromatography for purification of 125I-labeled human prolactin.
We assessed a simple method for purifying 125I-labeled human prolactin, taking advantage of the abundant supplies of monoclonal antibodies available. 125I-Labeled human prolactin purified by immunoaffinity chromatography is compared with that purified by gel filtration on Sephadex G-100. We used monoclonal antibodies to prolactin to prepare the affinity chromatography columns. Prolactin was rad...
متن کاملPlurihormonal cells of normal anterior pituitary: Facts and conclusions
INTRODUCTION plurihormonality of pituitary adenomas is an ability of adenoma cells to produce more than one hormone. After the immunohistochemical analysis had become a routine part of the morphological study, a great number of adenomas appeared to be multihormonal in actual practice. We hypothesize that the same cells of a normal pituitary gland releases several hormones simultaneously. OBJE...
متن کاملDo serum prolactin levels correlate with antibodies against desmoglein in pemphigus vulgaris?
BACKGROUND Pemphigus vulgaris is a chronic inflammatory disease of skin, the etiology of which is not completely known. Despite the latter, anti-desmoglein antibodies play a proven role in the pathogenesis. Recent studies showed an etiologic effect for prolactin in the pemphigus vulgaris. This study aimed to quantify the correlation between serum prolactin levels and anti-desmoglein antibodies ...
متن کاملDoes Prolactin Indicate Severity of Psoriasis?
Background: Psoriasis vulgaris is a common chronic and inflammatory disease of skin that its etiology is not completely known. However, it seems that genetic and environmental factors play a role in this disease. In vitro studies show that prolactin as a neurohormone has an etiologic effect on psoriasis. This study aimed to compare serum prolactin levels in psoriatic patients with control group...
متن کاملSerum prolactin level in psoriasis
The recent report, “Serum prolactin level in psoriasis”, is very interesting 1. Ghiasi et al. concluded that “it seems that while intracutaneous prolactin may contribute to the pathogenesis of psoriasis, the probable role of pituitary-produced circulating prolactin in the disease process needs further investigation .” There are some concerns on this report. First, although Ghiasi et al. have st...
